The primers used for nested PCR were the sense primer 5′-GTTCCAGCAACTGCAAACAGTAGCAATG-3′ and antisense primer 5′-ACCTTTCTGCTTCTTCATTTAATTTCCC-3′. The PCR product from the first reaction was used as a template for nested PCR. The conditions were 37 cycles of denaturation at 94☌ for 2 min, annealing at 60☌ for 10 s, and polymerization at 72☌ for 30 s. Briefly, two sets of primers were used: the first PCR sense primer 5′-CAAGCAGCAGGAGGGAGAAGCTG-3′ and antisense primer 5′-TCCTACCCCCATAATTTGATCCAC-3′ were used to amplify a 296-bp PCR product. Nested PCR was performed as described previously ( 15). DNA extraction from joint fluids was performed using a G-spin™ Genomic DNA Extraction Kit (iNtRON, Korea). Total DNA was extracted using a G-DEX™ Genomic DNA Extraction Kit for blood (iNtRON, Korea) following the manufacturer's manual. The cell pellet was resuspended in 0.5 ml of isotonic phosphate-buffered saline solution. The cell pellet was resuspended in 5 ml of isotonic phosphate-buffered saline solution or appropriate cell culture medium and mixed by gently pipetting in and out. After washing, the cells were centrifuged at 250 × g for 10 min. After centrifugation, the interface was carefully transferred into a clean conical centrifuge tube. Centrifugation was performed at 400 × g for exactly 30 min at room temperature. Briefly, 3 ml of leukocytes was placed onto Histopaque®1077. Peripheral blood mononuclear cells (PBMCs) were separated using the Ficoll gradient centrifugation method using Histopaque®1077-1 (Sigma Aldrich, USA). We collected leukocyte samples from all goats in the farm through the jugular vein and extracted total DNA from peripheral blood lymphocytes (PBLs) for CAEV detection by PCR. Perhaps because of nervous system signs, nervous system symptoms such as lameness, body paralysis, tremor, and torticollis were found ( Figures 1A,B). In young kids (under 3–4 months old), the most common symptoms were pneumonia and nervous system signs. Farmers reported that the goats were found dead mostly within 1 month or after a maximum of 3–4 months after clinical signs started. Two adults older than 1 year had mastitis, arthritis, and a rapid drop in milk production. As shown in Figures 1A,B, swelling of the carpal joints, stiffness, difficulty moving and standing, distortion of the leg, and cachexia were common in most goats suspected of being infected. Clinical signs were found in 11 adults older than 6 months and 10 young animals between 6 months and 1 year old. The farm was isolated from villages in mountain areas. With careful observation, we found that more than 60 adult goats showed similar clinical signs. In particular, one Saanen dairy goat farm in Korea reported that some goats showed clinical signs such as arthritis, paralysis, swelling of the carpal joints, and even death. We previously published only genetic sequence information of CAEV in Korea, and we report our findings of clinical and pathological results in this report.Īmong the 111 goats tested, 56 goats were positive by PCR. Although there have been many similar reports, as there are a very small number of dairy goats in Korea, CAE reports are very rare. The aim of our study was to investigate the clinical and pathological findings of caprine arthritis encephalitis (CAE) in dairy goats in Korea. Latent infection is common to most lentiviruses, similar to CAEV, for which the infection remains latent until monocytes mature into macrophages ( 1, 14). Sexual transmission is also another suspected route of transmission, and some sources speculate that in utero transmission occurs however, additional studies and confirmation are still required ( 7, 13). There is horizontal transmission by direct contact between goats through prolonged contact between infected and healthy animals housed in the same location ( 12). After ingestion in milk, the virus crosses the small intestine and infects monocytes and macrophages ( 1, 11). The major route of transmission is considered vertical transmission through colostrum or infected milk. Although it is a widespread disease in most goat-farming countries, little information is available on the CAEV infection status of Korean goats. The arthritic form is the most common and is generally observed in individuals aged 6 months and older ( 9, 10). After being first diagnosed in goats in 1974, it has been diagnosed worldwide ( 5– 8). Infection with CAEV is persistent and lifelong. There are four major clinical forms/presentations: arthritis, encephalitis, mastitis, and interstitial pneumonia ( 1– 4). Goats infected with CAEV exhibit severe and chronic devastating disease characterized by significant economic loss. Caprine arthritis encephalitis virus (CAEV) is a single-stranded, icosahedral RNA virus of the family Retroviridae and belongs to subgroup B of small ruminant lentiviruses (SRLVs). This one is typically used when you want to deny the truth of something. ¿Quieres un café? – No, no gracias : Would you like some coffee? – No, no thanks. You could also throw in a “ gracias ”, depending on the situation. By repeating the words, your rejection doesn’t come off as impolite. No, noĪnother way to make something more clear is by repeating the word and saying “ No, no… ” You can use this one often when someone asks if you want something. If you want to be a little more polite when someone asks you a question, you can always increase the pitch of your voice or elongate the last sound: “ Nooo ”. The easiest way to say no in Spanish is just, “ no ”. Of course, this is the most obvious part. With all of these, your tone and body language help your listener know if you’re simply responding to them or if there’s some animosity behind the content. These ones are relatively neutral because what’s most important is the way that you say these. This first section will give you all the different ways you can say no in Spanish. Direct, but neutral ways to say no in Spanish So that’s why in today’s article, we’re going to be improving your Spanish vocabulary and giving you a ton of extra tools to respond to something negatively, without offending the other person. If all you do is give a negative answer, “no”, you could come off as a little rude. This one little negative word comes in handy a lot, but sometimes there are better ways to express ourselves in a certain context. This often happens when we want to say no in Spanish. One of the biggest problems we have as language learners is our lack of vocabulary we always end up using the same words over and over because we don’t know any other ways to express what we want. When you become conscious of what your organs do for you, take time to create a loving relationship with them. Use this technique to transform negative energies into positive ones. Simple practices, such as inner smile meditation, help you maintain and sustain health and wellness over the long-term. Learn more about what prana is and how to feel it. Read more: In Hindu, Ayurvedic, Hatha Yoga and Tantric Yoga traditions, this life force is called prana. When these negative emotions run wild, our health and vital life-force (qi) are compromised. Unfortunately, as we encounter stress in daily life, negative emotions-such as fear, anger, sadness, worry, and impatience-increase. Finding harmony within yourself cultivates contentment. Connecting with this inner friend helps you feel grounded, settled, and stable no matter what else goes on around you. When you smile to yourself, you become your own best friend. Smiling & Healing EnergiesĮarly Taoist texts suggest that maintaining a constant inner smile ensured health, happiness, and long life. Read more: Overcome the common habit of focusing on the negative over the positive with these exercises for negativity bias. Frowning activates hormones that increase blood pressure, weaken the immune system, increase susceptibility to infections, and exacerbate depression and anxiety. Smiling activates hormones that maintain blood pressure, improve respiration, accelerate healing, stabilize the mood, reduce pain, and relax muscles.Ĭonversely, frowning may increase stress and stress-related hormones. Research suggests that smiling reduces the stress hormones adrenaline, noradrenaline, and cortisol. The powerful psychological and physiological effects of this practice cascade through the body and improve physical and mental health, and awaken compassion. They also knew, more importantly, how the energy of the smile, when turned inward, can help us heal ourselves.Īn ancient Taoist technique that helps us stay grounded in the present moment and provides a burst of positive healing energy is often referred to as the inner smile meditation. They knew how the eyes transmit this energetic force outward to others. In ancient times, Taoists knew about the power of healing love generated from a smile. Mark Bertin reminds us how to practice joy and gratitude in difficult and stressful times. Read more: Life is full of both joys and sorrows. It may not feel all that balanced between the two, in part because of the relentless influence of our negativity bias. Imagine the effect of this emotional and energetic shift on your entire body, including your organs. Notice the energetic and emotional shift. Now frown for a moment furrow your brow and clench your jaw. Think of someone or something that brings joy to your heart. Intentionally separate your eyebrows to smooth your forehead. Relax your facial muscles, and then gently raise the corners of your lips. Put an easy smile on your face for a minute. And if you’re frowning, take a moment to smile instead. One way to stay balanced is by noticing throughout the day whether you’re smiling or frowning. Maintaining an emotional and energetic balance while you navigate daily obligations is essential. Life regularly offers both exhilaration and exhaustion. When done consciously, smiling can be literally healing. When we radiate a genuine smile, it opens our heart chakra and stimulates warmth deep within us. It offers positive energy and speaks directly to the hearts of others. Smiling is a universal language that extends compassion to others, without the need or use of words. When we extend love and kindness, which can be as simple as smiling, we not only help others feel loved and cared for, we feel a sense of inner peace and contentment as well. Have you ever noticed the effect of a smile? When we smile, joy and wellbeing radiate from our eyes and connects us with others, regardless of spoken language. The Easy But Powerful Way Of Inner Smile Meditation Insight Timer teacher Amy Patte Colvin explains the ancient Taoist practice of inner smile meditation. Generating high-quality life tables for long-lived species living in the wild is challenging for various reasons, with the long-term commitment to individual-based monitoring foremost among them. Specifically, age-specific survivorship l x and fertility m x, (where x indicates a discrete age class) are all that are needed to estimate the relative contribution of an age x individual to the future size of the population (the ‘reproductive value’, v x), the life expectancy of an age x individual ( e x), the average lifetime reproductive success (the average lifetime number of daughters per mother, R 0), the asymptotic population growth rate ( λ), the stable fraction of individuals of age x in a steadily growing or declining population (the age x entry in the ‘stable age distribution’, w x), and the average number of years between a female’s birth and the birth of her offspring (the ‘generation time’, G). The life table has a long history as a means of summarizing census and birth data to describe how survival and fertility vary with age, and of using these underlying parameters to estimate useful population-level statistics 1. These data have high potential for reuse they derive from continuous population monitoring of long-lived organisms and will be invaluable for addressing questions about comparative demography, primate conservation and human evolution. We used bootstrapping to place confidence intervals on life-table summary metrics ( R 0, the net reproductive rate λ, the population growth rate and G, the generation time). We also calculated reproductive value, life expectancy, and mortality hazards for females. In all species, our survival estimates for the dispersing sex are affected by heavy censoring. Using one-year age-class intervals, we computed point estimates of age-specific survival for both sexes. We provide male and female census count data, age-specific survivorship, and female age-specific fertility estimates for populations of seven wild primates that have been continuously monitored for at least 29 years: sifaka ( Propithecus verreauxi) in Madagascar muriqui ( Brachyteles hypoxanthus) in Brazil capuchin ( Cebus capucinus) in Costa Rica baboon ( Papio cynocephalus) and blue monkey ( Cercopithecus mitis) in Kenya chimpanzee ( Pan troglodytes) in Tanzania and gorilla ( Gorilla beringei) in Rwanda. Make a spore print form the tallest and fattest mushroom and you will have a good specimen for further research. When the mushrooms (fruits of the mycelium) start to grow and mature, they can be picked and their spores can be harvest for preservation. Wait until the whole substrate is colonized by the mushrooms mycelium before you start the fruiting process. Just drop a few milliliter in every corner of the mushroom substrate and you are ready to go.Īfter a few days white patches of mycelium will start growing on the places were the spores were dropped. A syringe contains 20 ml of spore solution, enough to inoculate 6-7 liters of substrate. With the mushroom spores syringe you will only need to drop 3 milliliter of spore solution to colonize 1 liter or 1.05 quart of substrate. Edible, medicinal or psilocybe mushroom strains need different substrates to grow on. This can be done by inoculating a substrate, a soil with nutritions specially developed for growing mushrooms. If you want to reproduce spores it will be needed to fruit some mushrooms from the chosen mushroom spores. The inoculation of substrate with spores syringe Have a look at the shape, size and color of the spores and what kind you have. Take a cover glass and put this on the drop of spores. Shake the spore syringe and add one drop of spore solution on to the microscope slide. Get a microscope slide and clean it properly. The spore syringe is now ready for further use, like microscopic research or ready to inoculate a sterile substrate. Before use shake the spore syringe for a few seconds. Remove the needle from it's sterile package (leave the protection cap on the needle) and attach it to the spore syringe. Unscrew the the cap from the spore syringe by turning it counter clockwise. Hold the syringe with the cap pointing up. Take spore syringe with the cap still on it. The syringe is sealed with a cap, called luer lock. 1x special spore solution in a 20ml syringe. Our mushroom spore syringe comes in a pack that includes: When the spore syringe was stored in the refrigerator it is best to take it out 12 hours before use. The concept of revenge bedtime procrastination itself is currently largely informal, in the sense that there’s no research that investigates it directly. This concept and similar ones are sometimes referred to using other terms, such as revenge sleep procrastination, sleep revenge, and revenge insomnia. “Learned a very relatable term today: “報復性熬夜” (revenge bedtime procrastination), a phenomenon in which people who don’t have much control over their daytime life refuse to sleep early in order to regain some sense of freedom during late night hours.” The concept of revenge bedtime procrastination was popularized in the following tweet, though the term has also been used previously by others: How to avoid revenge bedtime procrastinationĮxamples of revenge bedtime procrastinationĪn example of revenge bedtime procrastination is someone who spends almost all of their days either at work or dealing with errands, so they stay up late at night browsing social media and watching videos on their phone instead of sleeping, so they can feel that they have some control of their schedule.Īnother example of revenge bedtime procrastination is a teenager who spends almost all of their days doing what their teachers and parents tell them to do, so they delay going to sleep in secret because that’s a way for them to rebel and feel in control.Psychology and causes of revenge bedtime procrastination.Dangers of revenge bedtime procrastination.
